Scratch test
Cell scratches experimental service principle
Cell culture, observe the migration of cells to non-cell scratch regions to determine the migration capability of the cell
Experimental service purposes
Cell scratch method Detection of cellular migration capabilities
Experimental instrument
ultra-clean workbench, CO2 incubator, biological inverted microscope, 6-well plate (other can, but feel 6 holes, good, size),
marker Pen, straight ruler 20, microlifier head (sterilization), serum medium, PBS
experimental preparation
All sterilized instruments must be sterilized, ruled and Marker pen in UV irradiation before operation 30min (Ultra-clean station)
Experimental process
1. First use the Marker pen behind the 6-well plate, with a straight comparison, uniformly scratched line, about 0.5 ~ 1 cm- Tao, cross the hole. At least 5 lines per well
.
2 is added to about 5 x 105 cells in the air, and the specific quantity is different due to cells, and the mastery is overnight.
3. The next day with the gun head is compared to the straight ruler, try to be on the horizontal scratches behind, the gun head is vertical and cannot be tilted.
4. Wash the cells 3 times with PBS, go to the cells, and serum-free medium.
5. Put 37 degree 5% CO2 incubator, cultured. Press 0,6, 12, 24 hours sampling, taking photos.
1, use the Marker pen behind the 6-well plate, evenly spread horizontally, approximately 0.5-1 cm- channel, cross-passing. At least 5 pieces of
2 per well, about 5 x 105 cells are added to the well, and the specific quantity is different due to cells, and the mastery is overnight.
3 The next day, I used to take the head than the ruler, and I think it is necessary to take the trapping of the trapping behind and take the head to be vertical.
4, use PBS to wash cells 3 times, removed cells, and serum-free medium.
5, put in 37 degree 5% C02 incubator, cultured. Press 0, 6, 12, 24 hours to sample, take pictures.
